How about talking about the boy who served as a basis for Atom in the series?
Btw, the scene of Tobio's death from the original mangá was reproduced in the game Tetsuwan Atom: Heart no Himitsu.
Btw, would you be mad if they did Atom as his robot brother?
With this we could imagine several situations like Tobio acting as a actual brother for a robot which share some of his traits or being a boy who hates the robot.
Also it would interesting if Tobio has also meet Uran and specially how he would react about her.
I personally don't care about Cobalt, so I'm not commenting about him.
Uran is enough as a parent figure.

This is an interesting basis. Since Tenma was driven by his son's death, and later by the realization that the robot couldn't replace the boy, my first thought was that they would simply live as an ordinary family, which would change a bit Astro's destiny; but since you basically replaced the conflict Tenma felt with another one coming from Tobio, it could also become a trigger for the action.

I was thinking about Tobio defending the robots rights because he had a "brother robot" and then a "sister robot".
As for a theme which the "human brother" hates and wishes to destroy the "brother robot" I would use Rock Holmes or "Rokkuro" as he also is called.

HUGS- ASTROBOY would be a sweet brother robot!

I would not like to make Tobio so revolted about a robot copy, so it would be nice if he was a very compassionate brother instead.
But, I thought about the father of Rock Holmes making a replica of himself based in his past which he was noble and righteous.
So, the conflict would between Rock and his robot replica for example.
Also, it would be funny having Tobio reminding Uran that he is not a robot, lol
